Darkest Knight fought and beat Perpetua. Perpetua, is an entity from a race called The Hands. In the DC continuity, The Hands create, shape, and destroy Multiverses. Yeah, as in plural. The Darkest Knight defeated an entity that can destroy Multiverses. Meanwhile, The Spot hasn't shown anything close to that power. So, Darkest Knight all the way.

The Darkest Knight is an actual god while the Spot can only create portals (which is a slept on ability but not compared to an actual deity like TDK).

Spot to teleport him anywhere sure we still haven’t seen his full power but we did see a bit and it seems like he is a living black hole so I’ll give him gravitational manipulation which would make him over power also anything and any attacks are show to turn to dust and sucked into him so sorry spot
